Airflow delivered in parallel streamlines at a validated, uniform velocity through a HEPA or ULPA filter bank, continuously sweeping particles away from the critical zone in a single direction to maintain Grade A/ISO 5 conditions over exposed sterile product, open containers, and closures during aseptic processing.
Unlike turbulent (non-unidirectional) airflow, which relies on dilution to reduce particle concentration, unidirectional airflow physically displaces particles out of the critical zone before they can settle, which is why it is required wherever sterile product or components are exposed.
Airflow velocity, uniformity, and the absence of turbulence-generating obstructions — equipment, personnel movement, poorly designed fixtures — are all qualified together through velocity mapping and smoke (airflow visualization) studies, since an obstruction can locally disrupt the unidirectional pattern even where average velocity meets specification.
First air — air that has passed through the final HEPA filter with no intervening obstruction — must reach the critical exposure point directly, which is why operator and equipment positioning relative to unidirectional airflow is a routine aseptic technique assessment point.
