Moist heat is the most widely used and generally most robust terminal sterilization method because saturated steam transfers heat far more efficiently to microorganisms than dry heat, allowing lower temperatures and shorter cycle times for equivalent lethality.

Cycle design must account for the load’s heat penetration characteristics — a fully loaded, worst-case configuration is used during validation because air pockets or shielded surfaces can create cold spots that receive less lethality than the chamber probes indicate.

Where the product cannot tolerate terminal moist-heat sterilization, manufacturers must justify use of a lower sterility assurance method such as aseptic processing, per the decision hierarchy set out in EU GMP Annex 1.
