What a definition is not
A definition is SPEQ’s plain-language decode of how a term is used in practice, cited to the documents that define it. Depyrogenation is the process of removing or inactivating pyrogens — chiefly bacterial endotoxins — from product-contact components such as glass vials and stoppers. Because endotoxins are far more heat-stable than microorganisms, depyrogenation (typically dry heat) requires much higher temperatures or doses than sterilization.
Dry-heat depyrogenation of glassware is validated to achieve at least a 3-log (1000-fold) reduction of a known endotoxin challenge, commonly using an endotoxin indicator spiked onto or into representative containers. Typical tunnel/oven conditions run well above sterilization temperatures.
Not all components tolerate dry heat: elastomeric stoppers are usually rendered low-endotoxin by validated washing rather than dry-heat depyrogenation. The distinction matters because sterilizing an item does not depyrogenate it — endotoxin from dead cells persists.
